实验操作
To the mixture of 2-methylphenylboronic acid (30.4 kg, 224mol), 14 (28.1 kg, 102mol), and pyridine (32.2 kg, 407 mol) in DMF (169 L) was added anhydrous Cu(OAc)2 (2.77 kg, 15.3mol), and the batch was stirred for 37 h at 25 °C under an atmosphere of air; then HPLC analysis indicated <3% 14 remained (HPLC method A).

Acetonitrile (700 mL, 7 vol), copper(II) acetate (37.6 g, 0.5equiv), 2,2′-bipyridyl (32.3 g; 0.5 equiv), sodium carbonate (87.6 g, 2.0 equiv), 2 (100 g), and cyclopropylboronic acid(10.6 g, 0.3 equiv) were sequentially charged at rt under an atmosphere of 5% O2 in N2.
The resulting mixture was heated at 55 °C and vigorously agitated for 3 h while bubbling subsurface 5% O2 in N2 through the mixture. Four additional portions of CPBA (10.6 g, 0.3 equiv each) were charged every 3 to 4 h, and the progress of coupling was monitored by HPLC. Desired >98% conversion was typically reached after 16−24 h.
